The Mouse Insulin ELISA quantitates Ms Insulin in mouse serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Ms Insulin. Principle of the method The Mouse Insulin solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. Insulin ist ein Peptidhormon, das von Betazellen der Pankreasinseln sezerniert wird. Es reguliert den Kohlenhydrat-, Protein- und Lipidstoffwechsel durch die Verbesserung des Membrantransports von Glucose, Aminosäuren und bestimmten Ionen. Es fördert zudem die Glykogenspeicherung, die Bildung von Triglyceriden und die Synthese von Proteinen und Nukleinsäuren. Insulin ist ein 51-Aminosäure-Polypeptid-Produkt, das aus einem Vorläuferpeptid, dem Proinsulin, hergestellt wird. Proinsulin wird posttranslational in zwei Ketten (Peptid A und Peptid B) gespalten, die kovalent über zwei Disulfid-Bindungen und ein Molekül C-Peptid miteinander verbunden sind. Insulinmangel führt zu Diabetes mellitus, einer der Hauptursachen für Morbidität und Mortalität in der Allgemeinbevölkerung. Insulin ist auch bei Tumoren mit b-Zellenursprung wie Insulinom vorhanden.
